    In this study, the method of developing "syokuiku"( food and nutrition education) curriculum under clear attainment targets was clarified through practicing syokuiku program. Experience through cooking practice is very effective to achieve both class and personal targets. In addition, conscious observation about the changes of students and careful advices were important for the achievement.

    In this study, a curriculum of 'Shokuiku' (Japanese new food and nutrition education) with clear aim was proposed for special-needs school (mentally-retarded) and the effectiveness was verified through education practice. The former year tuition's contents of a special-needs school were clarified into an aim category of Shokuiku and the contents were rearranged to make a new curriculum for the year. Most of the school teachers appreciated that the new curriculum can be easily executed and the aim is clear. In senior high school section, the class including cooking practice to study food balance increased an incentive to learning and enabled most students to realize and understand the importance of food balance. In all school sections, each class or individual child set a learning goal and more than 50% of classes and children achieved the goals. These results indicated that the curriculum created in this study may serve as a model curriculum of Shokuiku for special-needs school.

    1. The curriculums for food education(' Shokuiku') were analyzed to clarify what was mainly learned at elementary schools specified for' Shokuiku' research school in Hyogo Prefecture.' Health and nutrition' was the mostly-studied category, and' importance of enjoying to eat',' building a good relation through having a meal together',' cooking' and' production of food' were also learned favorably. 2. Instruction plans of one hour class or through a year were collected mainly from research schools in Hyogo Prefecture, and brought together in the database where retrieval by keyword was possible. 3. Framework of the aims of food education, was provides and a tentative model curriculum was presented in the combination of all subjects, the period for integrated studies and special activities. An instruction plan was made from the model curriculum, and the validity of the model curriculum was confirmed by evaluating the activities in M elementary school according to the standards determined in agreement with the aims.

    江戸期の調味料である「いり酒」を料理本の記述をもとに再現し,現代への活用を検討するために,味や色に関わる成分分析や官能評価などから嗜好特性を明らかにすることを目的として研究を行った。いり酒は,古酒をベースに鰹節のイノシン酸ナトリウムのうま味と梅干のクエン酸の酸味が加わった調味料で,醤油よりも低塩分であり,冷蔵での保存が可能であること,また早いり酒は,野菜に使用すると好まれる傾向にあることから,現代の食生活に活用できる可能性が示唆された。

    1. Preincubation of L929 cells with various polyunsaturated fatty acids enriched the corresponding fatty acids in phospholipids, and some fatty acids attenuated cell death induced by TNF. The order of effective PUFA activity was DHA>DPA(n-3)? EPA>AA = 20:3(n-6)? 22:4(n-6). DNA laddering was not detected, and cells were coincidently stained with both annexin V-FITC and propidium iodide, indicating that the death mode was necrotic. Combined with our previous data (J. Nutr. 130: 1095-1101, 2000), DHA-enriched membranes can protect cell against TNF irrespective of death modes and that membranous DHA may abrogate the death signaling common to necrosis and apoptosis. 2. Various antioxidants such as N-acetyl cystein (NAC), 2-mercaptoetnanol (2-ME) reduced the apoptotic cell perventages induced by TNF in dose dependent fashion. Water-solible antioxidants increased cellular glutathion level, but inhibitory activity of antioxidants against TNF-induced apoptosis did not correlate with the glutathion-increasing activity. It was indicated that cellular glutathion level shows only little effect on cellular sensitivity to apoptotic stimulation by TNF, and that antioxidants can inhibit TNF-induced apoptosis by directly scavenging reactive oxygen species, constructing a part of apoptotic signals of TNF. 3. The effect of fatty acid supplementation on death of PC12 cells induced by methyl mercury. Different from apoptosis or necrosis induced by THF, supplementation of DHA or arachidonic acids in cell culture accelerate methyl mercury-induced cell death. This indicates that methyl mercury induces cell death.

    Step Up Education Program on Food, Nutrition, Cooking, Healthy Intake and Culture By Internet in Lifetime The Purpose of this project is to propose education programs on food, nutrition, cooking, healthy intake and culture by internet in lifetime. It was found that the cooking skills of younger generation and middle-aged men were very low. Individual differences were also found in cooking skill and eating habits. It was necessary to make step up programs on cooking, food and nutrition for good health in lifetime. This program has buffet-styled structures in every course on internet. It has four Levels for studying and we can select them at any time on demand. The content of this Education Program was on http://www.geocities.jp/syokuedu/.

    マリファナの主成分であるΔ9THCに対する受容体の内因性リガンドN-arachidonoyl-ethanolamide(アナンダミド anandamide、AEtA)、2-arachidonoylglycerol(2-AG)の量的変動を知り、さらには種々の精神疾患・行動異常に対する脂肪酸栄養の効果とアナンダミド産生の関連を明らかにすることを最終目標として、本研究を開始した。 以下に研究成果を記載する。 1.同一サンプルからのAEtA、2-AGの分離定量法の検討;両内因性リガンドとも、脂質抽出画分から、TCL-HPLC(GLC)により分離定量できるが、TLC上では、ほぼ同一の位置に分離されるため、同一サンプルから同時に両者を定量することは困難であった。しかしながら、内部標準物質、TLC展開溶媒を工夫することにより、2次元TLC上で両者を初めて分離することに成功した。 2.ラット食餌脂肪酸によるΔ9THC受容体内因性リガンドレベルの変化;ラットに次のような食餌を与え、2週間飼育した後の腎臓及び脳のAEtA、2-AG量を調べた。A群(コントロール);無脂肪食+ラード10%、B群;無脂肪食+リン脂質型DHA10%、C群;無脂肪食+トリグリセリド型DHA10%、D群;トリグセリド型リノール酸10%、その結果、BCD群ともに各臓器中のリン脂質の脂肪酸レベルにおいては、食事中の脂肪酸構成に見合った変化がみられたが、アラキドノイルグリセロールにはほとんど変化がみられなかった。しかし、脳のAEtAは、DHA食(B、C群)を与えることで低下した。 3.アナンダミドの分離において、TLC上の非常に近い位置に、細胞内のシグナル伝達に関わる脂質メディエーターの一つと考えられている。セラミドが分離されてきた。このセラミドについて、微量定量を目指し、種々検討した結果、同様の蛍光HPLCによる各分子種の分離定量法を確立することができた。 4.DHAによりTNF誘導のアポトーシスが抑制されることがわかった。

    1. Highly DHA-containing phospholipid (DHA-PL) was administered to rats and the ability to alter the fatty acid composition of the tissue lipids was compared with that of triglyceride (TG). The results indicate that n-3 fatty acid containing PL more efficiently decreases n-6/n-3 fatty acid ratio of serum and blood cells than TG. 2. The stability of didocosahexaenoyl-GPC (DHA-PC) against peroxidation was assessed in comparison with that of tridocosahexaenoate (DHA-TG). When DHA-PC was autoxidised in glass tubes without solvent, apparent generation of PL-hydoperoxide was approximately 1/100 of that of TG-hydoperoxide. However the half-life period of DHA-PL was less 1/10 of that of DHA-TG,indicating that DHA-PC was much less stable in oxidative degradation than DHA-TG,but that PC-hydroperoxide level was much less than that of TG-hydroperoxide. 3. Conclusion ; The stability of lipids against hydroperoxidation has been considered to be depend on the number of double bond of fatty acid residue. However our results indicate that rate of oxidative degradation of lipids also depends on the chemical form of fatty acid ester and that phospholipids as food shpuld be more carefully handled or stored to prevent oxidative degradation than TG.Also nutritional effects of PC form of n-3 fatty acids may be different from that of TG form and n-3 fatty acid-containing PL may be more useful to prevent thrombosis and related heart diseases than other chemical form of n-3 fatty acids.

    1.N-アセチルスフィンゴシン及びその類縁体の合成とアポトーシス誘導能について Heroldら(Helv. Chim. Acta 71,354,1988)及びMurakamiら(Tetrahedron Lett. 35,745,1994)の方法に準じ、スフィンゴシン=(2S,3R,4E)-2-amino-4-octadecene-1,3-diol及びフィトスフィンゴシンの全合成を行った。この方法はキラルなジアミンとスズを触媒とし、アルデヒド(スフィンゴシンの場合はTMS-アセチレンアルデヒド)とシリルエノールエーテルを縮合させる合成法であり、対掌性は完全に保存されている。スフィンゴシン、フィトスフィンゴシン及びスフィンゴシンのOH基を個別に保護したもの、2重結合を水添したもの、3重結合としたもの(反応中間体として得られる)、さらにはcis-trans異性体のN-アセチル誘導体について、ヒト白血病細胞株HL60細胞に対するアポトーシス誘導活性を調べた。活性測定は、顕微鏡による核の凝集や電気泳動でのDNAラダーの観察を指標に行った。その結果、種々のセラミド誘導体25μMを細胞に添加した時、16時間後に誘導されたアポトーシス細胞は、セラミドの膜透過型誘導体(C6セラミド,trans体)では約20%であったのに対し、cis体はその約2倍、3重結合を有するものは約4倍アポトーシスを誘導した。2重結合を水添したものでは、活性が認められなかった。アシル基にアセチル基を導入したC2セラミドでも同様の結果が認められ、3重結合体では有意にアポトーシス誘導能が増強した。アポトーシス誘導のメカニズムについては現在検討中である。 2.セラミドの微量定量法の開発について アンスロイリルニトリルを蛍光試薬として、セラミドのアルコール基と反応させ、HPLCで分離、定量する方法の確率を目指してる。これまでに内部標準になるC17セラミドを合成し、これに蛍光試薬を反応させる条件をほぼ確立したが、さらに高い回収率が得られる条件を検討中である。

    (1) We established a specific and highly sensitive method for the determination of MDA, which had been assumed to be an indicator of lipid peroxidation. (2) We made an apparatus to measure oxygen consumption, which was the most reliable index for the evaluation of lipid peroxidation. Based on the oxygen consumption, the efficiency and the limit of the parameters such as MDA, TBA-RS and tocopherols were in the autoxidation of soybean and rapeseed oils were investigated. (3) Liver regeneration taking place after the intoxication of carbon tetrachloride which damaged the liver by extensive lipid peroxidations was found to be regulated by alpha_1-receptor, which controlled liver regeneration following partial hepatectomy. (4) Ascorbic acid is known to be the most important anti-oxidant in biological systems. We established the method to determine the vitamin specifically and sensitively involving chemical derivatization and HPLC. (5) As a chemical index of aging, we developed a sensitive and specific method to determine peroxides and hydroperoxides using chemiluminescence. (6) The toxic mechanism of cadmium to the lung was investigated by evaluating the change in antioxidant system after the instillation of the metal into rat trachea. Based on the change of glutathione, it was found that cadmium caused lipid peroxidation.
